The Comparison of Th17andTreg Cytokines in Local and Systemic ImmuneResponses in Patients with Unexplained Recurrent Pregnancy Loss (URPL):New Insights in URPL

The balance between Th17/Treg in peripheral blood and reproductive tissues can play a role in unexplained recurrent pregnancy loss (URPL) pathogenesis. The aim of the present study was to evaluate the signature cytokine and Tregulatory marker (TGF-β and FOXP3) as well as the evaluation of the determinant cytokine of Th17cells(IL-17A) in patients with URPL and healthy women. Samples from 14 patients with URPL were taken as a case group and 12 normal no pregnant women as a control (normal) group collected from peripheral blood, endocervix, endometer and vagina. Gene expressions were measured by qRT-PCR. The serum and cervicovaginal fluid levels of cytokines were measured by ELISA.. IL-17 gene expression was significantly higher in peripheral blood in women with URPL compared to control group (P= 0.01). The cervicovaginal fluid TGF- β level in the case group and the normal group was different (P=0.015). This study indicated that dysregulation of systemic immune responses may reduce regulatory immune responses in reproductive tissues which may play a critical role in the pathogenesis of URPL.
