CARACTERIZACIÓN HISTOLÓGICA DE LA EMBRIOGÉNESIS SOMÁTICA A PARTIR DE LIMBOS FOLIARES DE BONIATO (Ipomoea batatas L. Lam.)

2005
With the use of the culture medium proposed by Murashige and Skoog, supplemented with myoinositol (100 mg.L -1 ), thiamin (1 mg.L -1 ), MS vitamin (10.0 ml.L -1 ), sucrose (3 %), gelrite (0.2 %) and different growth regulators in the culture medium, depending on the experimental objective, the callogenesis and somatic embrygenesis were described in explants from sweet potato leaves of INIVIT B 93-1 cv, through a callus histological study; the site where cell proliferation provided the induction of a endogenous callus was determined. The callus could be seen after six days from the aseptic inoculation. At the beginning, the callus was formed from vascular bundles and later, due to the intense mitotic activity, it began to occupy the whole mesophyll. This resulted in a compact mass, destroying the explant epidermis and giving birth to a heterogeneous yellow nodular mass outwards, and five days later, appearing the embryogenic primordia that led to somatic embryos after 20 days.
