Effects of Different in vitro Pollination Methods on Capsule Formation in Lilium Hybridization

To overcome incompatibility caused by pre-fertilization barriers in lily breeding,cultivars from five series of Lilium hybrids and one wild species as experimental materials,different in vitro pollination methods such as in vitro stigmatic pollination,in vitro cut-style pollination and in vitro ovarian pollination were used to carry out the hybridization.The results showed that in intraspecific crosses,percentages of swollen ovaries and shaped capsules were 30 % and 15 % for in vitro stigmatic pollination,15 % and 5 % for in vitro cut-style pollination,25 % and 5 % for in vitro ovarian pollination.In interspecific crosses,percentages of swollen ovaries and shaped capsules for the three in vitro pollination methods were 17 % and 13 %,3 % and 3 %,10 % and 3 %,respectively.Hybridization type had a great effect on capsule formation;Percentages of swollen ovaries and shaped capsules both were higher in intraspecific crosses than in interspecific crosses.In vitro stigmatic pollination had the best effect for overcoming the pre-fertilization barrier in lily hybridization;the other two methods showed less and same effectiveness.Hybrid seedlings of some combinations were obtained by virtue of embryo rescue and identification of the hybrids was accomplished with ISSR markers,which verified the truth to obtain lily hybrids of interspecific crosses using in vitro pollination technique.
