SPECTROPHOTOFLUOROMETRIC DETERMINATION OF LOW CONCENTRATIONS OF AMOBARBITAL IN PLASMA.

Abstract A method for the fluorometric determination of amobarbital in plasma is described. Following extraction of amobarbital from the plasma with butyl ether and subsequent washing of the butyl ether extract with pH 8.0 phosphate buffer, the ether layer is re-extracted with 1 M NaOH. The alkaline extract is then read on the Aminco-Bowman spectrophoto-fluorometer at 265 mμ excitation wavelength and 410 mμ fluorescence wavelength, slit arrangement No. 4. Amobarbital concentration is proportional to fluorescent intensity in the range 0 to 3.0 μg/ml.
