Antimicrobial activity of synthetic chalcone-derived molecules in mixed biofilms and in vitro and in vivo toxicity

The failures of the endodontic treatments are associated, in part, to the presence of a resistant microbiota, being Enterococcus faecalis and Candida albicans the main microorganisms found in the root canals. The objective of this work was to prospect 30 chalcones to determine the MIC and MFC / MBC, to verify the anti-biofilm activity of the best compound, as well as to use the G. mellonella model to study the toxicity of the molecule. Aminochalcone were synthesized and diluted in DMSO for the accomplishment of the MIC and MFC/MBC experiments and from the results of the MIC the molecule I-38 was chosen for biofilm studies. The effect of I-38 on the biofilm of E. faecalis and C. albicans was evaluated by the quantification of CFUs. aminochalcone I-38 presented high antifungal activity, with MIC and MFC/MBC of 15.6 ?g / ml and for E. faecalis of 7.8 / 15.6 ?g / ml, respectively. During the use of 1x MIC and 10x MIC in the inhibition of biofilm formation when compared to control there was reduction of biofilm in mono and mixed biofilms in formation and mature. While the I-38 toxicity test on G. mellonella showed low toxicity at the concentrations tested. The aminochalcone I-38 thus has activity against the pathogens studied with potential to be used as intra-channel medication in future studies.
