Сравнительный экономический анализ получения первичной культуры фибробластов различной степени зрелости для лечения ожогов

2010
In spite of comparable clinical efficiency, the fibroblasts of varying degrees of maturity differ in their functional characteristics and require differential approaches for cultivation. So economic aspect of allogenic diploid fibroblasts (ADF) of varying degrees of maturity growing has been researched. Forming of monolayer culture of primary fetal fibroblasts (FF) has occurred approximately in 5.74 times quicker than mature skin fibroblasts (MSF) in the standard growing protocol. For the purpose of ADF growing protocol optimization an attempt of the most expensive component, fetal calf serum (FCS), volume reduction and its replacement by bovine serum (BS) has been realized. It has been revealed that the serum concentration decrease in the cultural medium has been crucial for the FF proliferation, but a source of the serum has not been of great importance. At the same time the attempt to replace FCS in part or in full by BS has led to interruption of the MSF proliferation as opposed to the FF. For long-term fibroblast ability to proliferate in conditions of subcultivation in vitro at the different variants of cultural media have been evaluated. The optimal for maintenance of the FF proliferation was the medium with 10% of BS, but for the mature fibroblasts with 10% of FCS. Herewith a first cost of the FF primary culture raising has turned out to be in 12.844 times cheaper then MSF, and the maintenance of the FF is cheaper in 2.77 times as opposed to MSF.
