Type 1 diabetic susceptibility encode by HLA, CTLA-4 and insulin gene in Brazilians subjects

Aims: To analyze the effect of HLA, CTLA-4 and promoter region of insulin gene on T1DM in Brazilians and would be to find markers that would best discriminate diabetic and non diabetic. Methods: “Time-resolved fluorometer” –1420 VICTORTM after hybridization with labeled probes - Eu(III)/Sm(III) and Tb(III. Results: The main predisposition alleles are DQB1*0302 (RR=3.84) and DQA1*03 (RR=3.0). There are a significant protecting effect of the DQB1*0301 allele. Analyzing only the DQA1 marker, we were able to correctly characterize between diabetic and non-diabetic in 71.13% of the analyzed individuals. When we added the DQB1 marker, this percentage increased to 82.47%. Conclusion: The main predisposition alleles were DQB1*02 (RR=4.66), DQB1*0302 (RR=3.84) and DQA1*03 (RR=3.0). According to the discriminant factorial analysis performed, the markers which best characterize and discriminate the two studied groups (diabetic and non diabetic) were DQA1 and DQB1.
