Method for generating surface cluster

The present invention relates to a method of generating a surface cluster comprising: amplifying the target sequence by a first universal primer and a second universal primer, wherein the first universal primer comprises a priming segment and the second universal primer comprises a priming segment, a modifier segment, and a marker segment, to produce a PCR product comprising a single-stranded tag, wherein the PCR product comprises a first strand and a second strand, and the second strand is attached to the single-stranded tag; providing a substrate, wherein the substrate comprises probes, first surface primers and second surface primers; applying the PCR products and probes to the substrate, thereby hybridizing the PCR products, the probes and probes to produce PCR product/probe complexes on the surface of the substrate; connecting the PCR product with a probe; rinsing the substrate; and extending the first surface primer and the second surface primer to finally form the surface cluster. In the method of the invention, the cluster density is controlled by the density of probes immobilized during substrate surface preparation by the substrate manufacturer or the ratio of surface primers to probes.
