Study on the anti-cerebral ischemia effect of Japanese pagodatree bark flavone in mice

Objective To study the effects of Japanese pagodatree bark flavone on cerebral ischemia in mouse. Methods The model of SD rats' arteriovenous shut thrombosis was produced by a polyethylene catheter with silk inserted into the right carotid artery and connected to the left external jugular vein. Kunming mice were put in a wide-mouthed and closed bottle in normal atmospheric pressure, and the surviving time of mice in normo-baric hypdxia was recorded. The model of cerebral ischemia in mice was Wished by ligating both common catotid arteries( CGA) ,and the effect of Japanese pagodatree bark flavone on surviving time of the mice was observed. The model of cerebral ischemia was produced by Ligalion of Bilateral CCA. After ischemia for 60 min followed by wet brain weight dry brain weight and water content of brain tissue was determined respectively, and the effect of Japanese pagodatree bark flavone on encephal edema was observed. Results In contrast with normal saline group, Japanese pagodatree bark flavone group can obviously inhibit the formation of thrombosis in arteriovenous shut in rats[ (0.0131 ±0.0011)g vs (0.0203 ±0.0009)g, P<0.01 ] prolonged the surviving time of mice in normobar-ic hypoxia[ (25. 81 ± 1. 37)min vs (18. 11 ±0.48)min, P<0. 01] ;increased the surviving time of mice ligating both CCA[ (380.20 ±16.02)min vs (273. 10 ± 12. 16) min, P<0. 01 ];reduced wet brain weigh[(0. 3350 ± 0.0281) g vs (0. 3617 ± 0. 0215 ) g, P < 0. 05 ] and water content of brain tissue [ (0. 2441 ± 0. 0220) g vs (0.2716 ±0.0157)g, P<0.05]. Conclusion Japanese pagodatree bark flavone has the protective effect on cerebral ischemia in mice. Key words: Japanese pagodatree bark flavone; Brain ischemia; Thrombosis; Surviving time
