TAVUK ETLERİ VE TAVUKLARDAN İZOLE EDİLEN SALMONELLA ENTERİCA SUBSP. ENTERİCA SEROVAR ENTERİTİDİS SUŞLARININ FAJ TİPLENDİRİLMESİ VE PULSED-FİELD GEL ELECTROPHORESİS İLE ANALİZİ

OZET Bu calisma tavuk ve tavuk etlerinden izole edilen Salmonella enterica subsp. enterica serovar Enteritidis ( S. Enteritidis ) suslarinin Pulsed-Field Gel Electrophoresis (PFGE) ve Faj tiplendirme yontemleriyle tiplendirilmesi amaciyla yapildi. Toplam 38 S. Enteritidis susu XbaI restriksiyon enzimi kullanilarak PFGE yontemiyle tiplendirildi. Tiplendirme sonucunda 7 farkli bant profili elde edildi. Analiz edilen 38 S. Enteritidis susunun 27 sinde (%71) ayni PFGE bant profili (XI) saptandi. Tum suslar arasindaki genetik benzerlik % 85 bulundu. Faj tiplendirme sonucunda 38 susun 15'i (%39.4) S. Enteritidis PT4(Faj Tip 4) olarak tiplendirildi. Suslarin  %18.4'i PT7, %13.1'i PT16, %10.5'i PT1, %7.8'i PT6 ve %2.6'si PT35 faj tipinde bulundu. Suslarin 3'unun faj tipi belirlenemedi. Farkli faj tipindeki (PT1, 4, 6 ve 7) suslarda ayni PFGE profili (X1) saptandi. Bu calismanin sonuclari Turkiye'de tavuk orijinli S . Enteritidis suslari arasinda bir genetik iliskinin oldugunu gostermektedir. Insan ve hayvan izolatlari arasindaki genetik iliskiyi ortaya koymak icin ilave calismalara ihtiyac duyulmaktadir. ENTERICA SEROVAR ENTERITIDIS STRAINS ISOLATED FROM CHICKENS AND CHICKEN MEAT BY PULSED-FIELD GEL ELECTROPHORESIS AND PHAGE TYPING ABSTRACT The aim of this study was to analyse S. Enteritidis strains isolated from chickens and chicken meat by Pulsed-Field Gel Electrophoresis (PFGE) and phage typing. A total of 38 S. Enteritidis strains were analysed by PFGE after digestion of DNA with XbaI restriction enzyme. Seven different PFGE profiles were observed. A predominant type (X1) was found in 27 (71 %) of 38 S. Enteritidis strains. For all the strains, the genetic similarity was 85%. The most common phage type was PT4, which was found in 15 (39.4%) of the strains. Other phage types were PT7 (18.4%),  PT16 (13.1%), PT1(10.5%), PT6 (7.8%) and PT35 (2.6%). Three strains were RDNC (reacted but did not conform). Different phage types  such as PT1, 4, 6 and 7 belonged to the same PFGE pattern (X1). The results of this study shows that S. Enteritidis strains originated from chickens in Turkey are genetically related. Further studies should be performed to determine genetic relationships among animal and human isolates.
