Effect of puerarin on expression of matrix metalloproteinase-9 and matrix metalloproteinase tissue inhibitor-1 in THP-1 monocyte-derived macrophages induced by oxidized low density lipoprotein

Objective To investigate the effect of puerarin on expression of matrix metalloproteinase-9(MMP-9) and matrix metalloproteinase tissue inhibitor-1(TIMP-1) in THP-1 monocyte-derived macrophages induced by oxidized low density lipoprotein(oxLDL).Methods THP-1 monocyte-derived macrophages induced by ox-LDL were cultured with different concentrations of puerarin and the expressions of MMP-9 and TIMP-1 were analysed by RT-PCR and ELISA.ResultsPuerarin inhibited the expression of MMP-9 concentration-dependently,but had no effect on the expression of TIMP-1.Conclusion Puerarin can make atheroseclerotic plaque stable by regulating the expression of MMP-9 in the macrophages.
