Effect of paclitaxel on the expression of vascular' s fibroblast of matrix metalloproteinases -2 and a - smooth muscle actin

Objective To investigate the effect of paclitaxel on the expression of matrix metalloproteinases-2(MMP-2) and α-smooth muscle actin(α-SM-actin)of the fibroblast.Methods Adventitial fibroblast(AF) were prepared from rat aortas using explant technique and cultured for passages,the expressions of MMP-2 and α-SM-actin were determined by RT-PCR.Results The expression of MMP-2 in the paclitaxel group is significantly lower than that in the control group(P0.01).The expression of α-SM-actin in the paclitaxel group is significantly lower than that in the control group(P0.01).Paclitaxel can inhibit the expression of MMP-2 and α-SM-actin of the fibroblast,effectively inhibiting the phenotype transformation of AF,thus inhibiting the multiplication and migration of vascular adventitia.Conclusion Paclitaxel can prevent restenosis after percutaneous coronary intervention.
