Effects of resveratrol on proliferation and expression of matrix metalloproteinase-9 in gastric carcinoma cells

Objective To observe the effects of resveratrol on proliferation of human gastric carcinoma cell line BGC823 cells and expression of matrix metalloproteinase-9(MMP-9) in vitro so as to explore the mechanism of resveratrol's inhibiting the metastasis of BGC823 cells.Methods BGC823 cells were treated with different concentrations of resveratrol(10,50 and 100 μmol/L)for 24,48 and 72 h,respectively.The effects of resveratrol on proliferation of BGC823 cells were assessed with methyl thiazolyl tetrazolium.The expression of MMP-9 mRNA was determined by reverse transcription polymerase chain reaction while MMP-9 protein was identified by Western blot analysis.Results Resveratrol could inhibit the proliferation of BGC823 cells in dose-and time-dependent manners.Moreover,compared with those in control group,both MMP-9 mRNA and protein expressions were markedly reduced after resveratrol treatment(P0.05).Conclusion Resveratrol can inhibit the proliferation of BGC823 cells and down-regulate MMP-9 expression.Therefore,resveratrol may suppress the invasion and metastasis of gastric carcinoma.
