Changes of Genes Expression Profile in Liver Regenerated from Major Hepatectomy after Ischemic Preconditioning(IPC) in Rats

Objective The aim of the research was to study the gene expression profile in liver regenerated from major hepatectomy after ischemic preconditioning(IPC) in rats.Methods Sprague-dawley(SD) rats were subjected to 30 min of total hepatic ischemia after ischemic preconditioning(10 min of ischemia,followed by 10 min of reperusion) and 70% hepatectomy was performed just before reperfusion and at 4,24,48 h after operation the total RNA was isolated from liver tissue of rats.The differentially expressed genes during different stages of liver regeneration were detected through affmetrix RAT GeneArray 1.0 ST gene chip technique and their functions were analyzed.Results During the process of liver regeneration liver tissue with a total of 1103 differentially expressed genes related to metabolism related genes,mitosis,cell cycle regulation genes,inflammation-related genes,apoptosis-related genes,signal transduction-related genes,cytokine-related genes,growth factor genesand so on.There are 7 kinds of expression trends.Conclusions The genome-wide changes in the expression profile in liver regenerated from major hepatectomy after ischemic preconditioning(IPC)in rats is complicated.Analyzing the changes in the gene expression by DNA chips can reveal the mechanism and may identify some drug targets for the treatment.
