Control of excitation in the fluorescence microscope

Abstract In fluorescence microscopy image brightness and contrast and the rate of fading depend upon the intensity of illumination of the specimen. An iris diaphragm or neutral density filters may be used to reduce fluorescence excitation. Also the excitation band-width may be varied by using a broad band exciter filter with a set of interchangeable yellow glass filters at the lamphouse.
