Effect of Tri-ortho-cresyl Phosphate on Activity and Translocation of Protein Kinase C in SH-SY5Y Human Neuroblastoma Cells

Objective To study the effect of tri-ortho-cresyl phosphate(TOCP) on the activity and translocation of protein kinase C(PKC) in SH-SY5Y human neuroblastoma cells.Methods SH-SY5Y cells were exposed to TOCP at concentrations of 0,0.25,0.5 and 1.0 mmol/L for 12,24 or 48 hours.The PKC activity was detected by PKC kit.The membrane translocation of PKC in cells was determined by indirect immunofluorescence and was observed by inverted fluorescent microscope.Results The PKC in the SH-SY5Y cells treated by TOCP at 1.0 mmol/L for 24 h was the most active,and the translocation of PKC from cytoplasm to membrane was induced.Conclusions TOCP can activate PKC and induce PKC membrane translocation in SH-SY5Y cells.
