Improving De Novo Nanopore Sequencing by Stretching DNA

Nanopore DNA sequencing is an emerging sequencing technology capable of long read lengths with minimal sample prep and low overhead costs. We report improvement of the accuracy of de novo sequencing with biological nanopore MspA. We control the motion of the DNA within the pore using a DNA helicase and a variable voltage simultaneously, which allows us to identify enzyme missteps and resolve otherwise indistinguishable base sequences. This addresses the primary error modes limiting the accuracy of de novo nanopore sequencing.
