Effect of Herbal Compound 861 on Hepatocarcinogenesis Induced by Diethylnitrosamine in Rats

Objective:To observe the effect of Herbal Compound 861 to the hepatocarcinogenesis induced by diethylnitrosamine in rats.Methods: 250 male SD rats were divided into normal group,model group and intervention group.The model and intervention group were administered 1% DEN intragastically once a week,for 14 weeks.The normal group was given saline instead of DEN,and intervention group were fed with Cpd 861 every day until the end of study.Seven to ten rats of model and intervention group were killed at 2,3,5,8,10,12,14,18 weeks,respectively.Four control rats were also killed at the same time points.The remained rats were followed to the end of the experiment at 26 weeks.Histopathological analysis were done by HE,Masson's trichrome of paraffin sections.The degree of necrosis,fibrosis and oval cell proliferation were analyzed.Immunohistochemistry and Western-blot was employed to detect the content of GST-P.Results: Cpd 861 intervention group had fewer nodules on outer surface of liver,less perivenular zone necrosis(P0.01),fibrosis(P0.01) and oval cell proliferation(P0.05) than model group,and the level of GST-P protein were less in intervention group.Conclusion:Herbal compound 861 can reduce the severity of perivenular zone necrosis,fibrosis and oval cell proliferation induced by DEN,delay the emergence of liver cancer to some extent.
