Sterilization conditions for tissue culture of different explants derived from Perilla frutescens (Linn) Britt

Seeds,cotyledons,hypocotyls and leaves of Perilla frutescens(Linn) Britt were used as explants and ster-ilized with different times of alcohol,mercuric chloride and NaClO to discuss their effects on the pollution and growth of explants and obtain the optimal sterilization conditions.The results showed that the different sterilized times of alcohol,mercuric chloride and NaClO had obvious effects on different explants.According to the germination rate and pollution rate,and the induction and growth of callus,the best sterilization method for seed was 75% alcohol in 20 sec and 0.1% mercuric chloride in 10 min,which germination rate and pollution rate were 50% and 36%,respectively.The best steril-ization method for cotyledons and hypocotyls was 75% alcohol in 20 sec and 0.1% mercuric chloride in four min,and the pollution rate and induction rate of callus were 7%,8% and 40%,36%,respectively.The best sterilization method for leaves was 75% alcohol in 30 sec and 0.1% mercuric chloride in 7 min,and the pollution rate and induction rate of callus was 10% and 7%,respectively.These results could provide the basis on establishing the regeneration system and rapid propagation of Perilla frutescens(Linn) Britt.
