Objectives To study HMGB1 expression of cultured human umbilical vein endothelial cells induced by LPS and its mechanism. Methods Cultured HUVECs at passage 3 to 6 were used for experiment to measure HMGB1 expression (by ELISA) stimulated by 10 µg/L LPS at 6 h, 12 h, 24 h, 48 h. Those to be compared with which were inhibited by PDTC. Results HUVECs didn ’ t release HMGB1 without stimulation, but the expression of HMGB1 was highly dependent on the stimulation of LPS in manners of time. And PDTC could significantly decrease the HMGB1 expression. Conclusions The induced activation of NF-KB by LPS starts up HMGB1 expression, which may there by influence the pathogenesis of atherosclerosis by HMGB1. So, we find HMGB1 may be an important factor to influence atherosclerosis in patients with coronary heart disease of course, its function need more reaseach.
