Sulfation for Modification of Lycium Barbarum Polysaccharide and Its Inhibiting Effect to Growth of Hela Cells in Vitro

The modification conditions were optimized for the preparation of sulfated lycium barbarum polysaccharide(SLBP) by using chlorsulfonic acid-pyridine method by L9(3)4 orthogonal design.The optimized conditions were 1∶4 of chlorosulfonic acid-topyridine,reaction temperature of 80 ℃ and reaction time of 2 h.The content of sulfur in SLBP and the degree of substitution(DS) were achieved to be 17.23% and 1.935,respectively.The nature of SLBP was characterized by UV spectra and infrared spectra analysis,and the inhibiting effect of SLBP to growth of Hela cell in vitro was tested with MTT method.In the range of 25~200 mg/L,LBP had little inhibited activity towards the Hela cells,while SLBP inhibited the growth of Hela cells remarkably with 31.71% inhibitory ratio.The SLBP prepared by the proposed sulfated method has higher sulfur content and degree of substitution,and obviously inhibiting effect to growth of Hela cells.The inhibitory ratio is positive correlation with the substitution degree and the tested dosage.
