Effect of Arsenic Trioxide on the Growth and Cell Cycle of Human Gallbladder Carcinoma Cell

Objective:To observe the inhibitory effect of As_2O_3 on the growth of human gallbladder carcinoma in vivo and in vitro and the alteration of cell cycle.Methods:Human gallbladder carcinoma cells(GBC)were cultured with different concentra- tions of As_2O_3,the proliferative activity of cells was detected by MTT methods.and the cell cycle status were carried out by flow cytometry(FCM).Twenty BALB/c nude mice,inoculated with the GBC cells subcutaneously,were randomly divided into nature saline(NS)and As_2O_3 groups.0.2ml NS or 10mg As_2O_3/kg was injected biweekly into the abdominal cavity of every mouse in.NS group or As_2O_3 group for seven weeks.The tumor size was measured.Results:As_2O_3 may inhibit the growth of human gallbladder carcinoma in vivo and in vitro,and in a time-and dose-dependent manner in vitro.GBC cells treated with As_2 O_3 were arrested in G1 phase.Conclusion:As_2O_3 can significantly inhibit the growth of human gallbladder carcinoma in vivo and in vitro as well as induce the G1 phase cell cycle arrest in vitro.
