Increased isolation and identification accuracy for Salmonella by introduction of polyclonal antibody against Proteus mirabilis

2012
Proteus mirabilis is one of the false positive bacteria in Salmonella detection. To increase the isolation accuracy for Salmonella,we prepared polyclonal antisera. The antisera which was purified with protein G column was mixed with Proteus mirabilis and then analyzed by scanning electronic microscopy.Proteus mirabilis was destroyed after 2 hours incubation with its antibody in vitro. In addition, purified antibody was added into Rappaport's broth(MM) and selenite cysteine broth(SC) to evaluate the microbicidal effect for Proteus mirabilis.Comparison between modified and traditional methods was carried out with more than 100 clinical samples. The results showed that the modified method could eliminate the interference caused by Proteus mirabilis and increase the isolation accuracy of Salmonella.
