Simultaneous determination of residues of diclazuril and toltrazuril in chicken tissues by high performance liquid chromatography

A simple and rapid determination of anticoccidial drug residues,diclazuril and toltrazuril,in chicken tissues has been developed.Diclazuril and toltrazuril were extracted with acetonitrile from chicken liver,muscle,and kidney.The extract was rinsed with N-hexane saturated with acetonitrile and then evaporated.The residue was dissolved in 1.0mL of mobile phase.then determined by HPLC on reversed-phase C18 column with 0.05 mol/L phosphate acid/acetate buffer-acetonitrile(40∶60) as the mobile phase.At the linear correlation 0.01-5.0 mg/L,the mean recoveries(n=5) of diclazuril and toltrazuril spiked in chicken tissues were 92.0%-102.0%(CV 5.9%-12.2%) and 83.4%-89.0%(CV 6.9%-8.6%),respectively.The detection limits of diclazuril and toltrazuril were 0.005 and 0.004 mg/kg,respectively.This method meets the demand of residues determination of diclazuril and toltrazuril.
