Effect of Bilateral Injection of Aβ25-35 in Amygdala on Learning and Memory and Expressions of Bax and Bcl-2 in Cortex of Rat Dementia Models and Qingxin Kaiqiao Formula and Saponin Interventions

Objective:To study the effect of bilateral injection of beta-amyloid 25-35 in amygdala on learning and memory and the expressions of Bax and Bcl-2 in cortex of rat dementia models and Qingxin Kaiqiao Formula and saponin interventions.Methods:Animal test used comparison method.Forty male SD rats were randomly divided into five groups:the control group,the model group,the Aricept group,the Qingxin Kaiqiao Formula group and the saponin group,with 8 rats in each group.AD was induced by Aβ25-35 injecting into the bilateral amygdala to observe the spatial learning and memony ability,the expressions of Bax,Bcl-2,Aβ and GFAP by immunohistochemistry method.Results:Learning and memory abilities of Qingxin Kaiqiao Formula groups were improved,the expressions of Bax、Aβ and GFAP decreased,the expression of Bcl-2 increased in the cortex of rat dementia models,compared with comparison group,the difference was notable(P0.05~0.01).Conclusion:Qingxin Kaiqiao Formula may obviously improve learning and memory abilities of rats with AD by decreasing the expressions of Bax、Aβ and GFAP,increased the expression of Bcl-2 in the cortex.
