PURPOSE: To obtain a new DNA encoding a lymphotoxin, capable of providing a variant obtained by performing a loss, insertion or substitution of amino acids to an amino acid sequence of the lymphotoxin, and encoding a recombinant lymphotoxin enhanced in an antitumor activity. CONSTITUTION: This DNA encoding an objective recombinant lymphotoxin, consisting of nucleic acids not containing non-translatable intron sequences or a DNA encoding the other proteins, or not having a natural sequence, is obtained by culturing human peripheral lymphocytes in a medium containing phorboemyriatae acetate, staphylococcus enterotoxin B and thymosin α-1 for 48 hr while producing the lymphotoxin, concentrating by adsorbing the mRNA on an immobilized oligo dT, eluting for synthesizing a cDNA by using a reverse transcriptase, treating the cDNA with a restricting enzyme for inserting into a vector and further inducing a position specific mutation.
