The basic research about the treatment of rapamycin combined with cisplatin

Objective By the cormparision of the rapamycin combined with cisplatin with the rapamycin or cisplatin alone towards osteosarcoma' s proliferation and apoptosis,to provide new method in treating osteosarcoma in the clinical.Methods Bone sarcoma cell line were randomly divided into 4 groups:The blank group,positive medicine group 1,positive medicine group 2 and the treatment group.to add 20 mg/L rapamycin in the positive medicine group 1,to add 10 mg/L cisplatin In the positive medicine group 2,to add 20 mg/L rapamycin and 10 mg/L cisplatin in the treatment group,and to add the equal amount of normal saline in the blank group.Then detect the Inhibition rate of every group with MMT,andthe apoptosis rate with flow evtometrv.Results to mean ± standard deviation (（x） ±（s）) said.Results The inhibition rate and apoptosis rate of combination towards osteosareoma enhances obviously compared to the separate application,and the difference is statistically significant(P ＜ 0.05).The blank group:0.33 ± 0.02,positive medicine group 1:0.56 ± 0.05,positive medicine group 2:0.55 ± 0.03,The treatment group:0.95 ±0.01.The apoptosis rate of combination towards osteosarcoma enhances obviously compared to the separate application,and the difference is statistically significant(P ＜ 0.05).Conclusion The combination of rapamycin and cisplatin can Inhibit osteosarcoma cell proliferation and induce its apoptosis obviously,better than the separate application.then can trear the osteosarcoma better.
