An improved assisting method for genetic transformation via Agrobacterium tumefaciens

When the cell suspension of Agrobacterium tumefaciens was mixed and co-cultured with plant cells, the mixture was disposed using Tween 20 and negative pressure simultaneously. According to the transient expression of gus report gene in the callus treated, the effect of the disposition was compared with that of using Tween 20 or negative pressure only. The results showed that appropriate treatment could increase the transformation frequency and survival rate while over-treatment harmed plant cells and decreased the survival rate. Addition of Tween 20 while using negative pressure could decrease the harmness caused by the over-treatment.
