Competitive combination with metallothionein between metallothionein antibody and ions of cadmium and zinc.

[Objective] To discuss the mechanism of metallothionein antibody(MT-Ab)effecting cadmium induced kidney injury by exploring the competitive combination with metallothionein(MT)between MT-Ab and ions of cadmium and zinc.[Methods] Groups were divided according to different treatments.Every group contained 5 different dose subgroups,namely 0,5,10,20 and 40 mg/mL respectively.Competitive enzyme linked immuno-sorbent assay(ELISA)was adopted.The extent of competitive combination was determined by photospectrometry at 490 nm.[Results] The concentration of coating MT was 1 μg/mL,concentration of MT-Ab was 32 U.The optical density decreased as the concentration of ions of cadmium and zinc increased.[Conclusion] There was obvious competitive combination between MT-Ab and ion of cadmium and zinc.MT-Ab can dissociate CdMT into free Cd ion,so as to lead to cadmium induced kidney injury.
