Effect of D-glucosamine on expression of Bcl-2 and WTp53 proteins of cell line Eca-109

Objective:To observe the effect of D-glucosamine[2-(3-carboxy-1-oxopropyl)amino-2-deoxy-D-Glucose,COPADG] on cell proliferation and apoptosis and expression of Bcl-2 protein and WTp53 protein in human esophageal cancer Eca-109 cells and discuss the possible mechanisms.Methods:Eca-109 cells were cultured in vitro by using RPMI-1640 and calf serum,then were exposured to COPADG at 0.03mmol/L concentrations for different time.Cell growth inhibitory rate was detected by MTT colorimetric assay;The cell morphological changes were observed by inverted phase contrast microscopy.Expression of Bcl-2 protein and WTp53 protein was detected by using flow cytometry and apoptosis rate was analyzed by using Annexin V/PI fluorescence staining togeth er with flow cytometry.Results:COPADG could significantly inhibit the proliferation of Eca-109 cells and induce them to apoptosis,and also decrease expression of Bcl-2 protein but expression of WTp53 protein didn' t show any defferences.Conclusion:COPADG has significant antiproliferative and proapoptotic effect on human esophageal cancer cell line Eca-109,and downregulates expression of Bcl-2 protein,which may play a very important role in apoptosis of Eca-109 induced by COPADG.
