Comparison of radiation enteritis mouse models established at different doses

Objective To explore the optimum dose for the mouse radiation enteritis model by abdominal irradiation(AR).Methods 84 BALB/c female mice were randomly divided into three groups as control group,6 Gy group and 10 Gy group.For each group,mice were sacrificed at 3,7,14 and 21 days after single abdominal irradiation of Cobalt-60,respectively.The blood was sampled to detect the hemogram,the liver and renal function.5 cm sections of the terminal ileum were fixed in formalin for histopathological examination including the mean heisht of villous and the number of crypt.Small intestinal mucosal samples were homogenized for NO concentration analysis.Results In 10 Gy group,WBC and PLT decreased obviously,and the liver function was damaged.The length of villi and the number of crypt were significantly reduced in irradiated groups compared with control group,especially in 10 Gy group.Intestinal NO concentration decreased in 10 Gy group in 3 d.Conclusions To establish the mouse model of irradiated enteritis single 6 Gy60Co abdorminal irradiation is optimum.
