Conditions for identification of codeine phosphate and its biodegradation products in culture media of Rhodococcus rhodochrous IEGM 647 by gas chromatography-mass spectrometry using solid-phase extraction were developed. Analyses of compounds in the form of acetyl derivatives and native materials have shown that major biodegradation products of codeine phosphate by cells of R. rhodochrous IEGM 647 are codeinone, hydrocodone and 14-hydroxycodeinone. It is shown that the relative contents of codeine and its metabolites in Rhodococcus culture media should be estimated without the derivatization procedure. A scheme of the codeine phosphate biotransformation by R. rhodochrous IEGM 647 has been described. The use of this biodegrading strain can be useful to produce 14-hydroxycodeinone, an important intermediate in the synthesis of narcotic analgesics and antagonists.
