TRANSIENT INCREASE OF PRODUCTION IN FUSED RECOMBINANT CHO CELLS

ABSTRACT Cells from the FVIII producing cell line ADLA were stained with FITC-DA(1) and X-RITC respectively and fused by Polyethylene glycol (PEG). Flow Cytometry analysis documented the successful fusion. One week after fusion, the PEG fusion treated cells show a significantly elevated FVIII expression in COATEST(2) (detecting the FVIII activity), in comparison with the original cell line. Antibody staining of fixed cells also showed an elevated FVIII antigen concentration intracellularly, as detected by Flow Cytometry. We are currently investigating the mechanism behind the elevated protein expression.
