Changes and significance of intestinal clostridium in obese children

Objective To analyze the diversity of clostridium in simple obesity children aged 7-14 and the difference from that of normal individuals, and to explore children's simple obesity affecting intestinal advantage bacterium of the genus. Methods We have collected fecal samples of thirty 7 to 14 years old cases with simple obesity and 30 healthy children’s faeces samples of the age. All bacterial DNAs were extracted from the faeces, and then clostridium sDNA 16 species specific primers were used for PCR amplification and separation by DGGE. The obese children’s and healthy children’s intestinal clostridium molecular fingerprint, diversity, and similarity were analyzed. At the same time, the obesity and normal children's weight, height, BMI, blood pressure, blood lipid, leptin, and plasma adiponectin were measure to investigate the relationship between the obese children with the advantage of intestinal bacteria genera and its clinical significance. Results The obese children's BMI, SBP, TG, TC, and LEP were higher than the normal group’s (all P 0.05). The obesity group’s stripe number of clostridium DGGE profiles and H'sample index were lower than the normal group’s; the cluster analysis results showed that the similarities between the individuals in the obese group and between the two group were reduced. The S and H' indexes of DGGE profiles in the two clostridium groups, the normal group, and each obesity group were negatively correlated with BMI and LEP, and positively with ADP. Conclusion Human intestinal flora of clostridium changes is related to childhood obesity; the diversity and similarity of obese children's intestinal clostridium species are decreased; the diversity of obesity children's clostridium is associated with BMI, ADP, and LEP.
