The invention discloses a technical method for rapid sterile seedling propagation of psammosilene tunicoides with axillary buds and immature stems. The method comprises the following steps: shearing psammosilene tunicoides with axillary buds and immature stems as an explant, inducing callus by utilizing the explant, differentiating sterile psammosilene tunicoides seedlings by utilizing the callus, performing optimization selection on types, concentration and matching ratios of different plant growth regulators, screening the optimal matching formula of the growth regulator and additives of psammosilene tunicoides in each period according to application of the different natural plant additives and the influence on psammosilene tunicoides induction and a seedling induction effect, thereby constructing a rapid vegetative propagation system of psammosilene tunicoides with the immature stems. According to aseptic seedlings obtained through vegetative propagation, excellent inheritable characters of maternal plants can be maintained to the greatest degree, and aseptic seedlings are provided. By virtue of the technical method, wild psammosilene tunicoides can be prevented from being subjected to extinctive excavation, the propagation coefficient of psammosilene tunicoides seedlings is greatly improved, and thus the method has important significance for protecting the threatened plant, psammosilene tunicoides, and restoring and protecting biological diversity.
