Abstract 11297: S-Glutathionylation, One of the Oxidative Protein Modifications, Stimulates Adipogenesis and Obesity by Stabilizing C/EBP Beta Through Inhibition of SUMO E3 Ligase PIAS1 Interaction

Introduction: Reactive oxygen species (ROS) are increased during adipogenesis and obesity. On the other hand, ROS stimulate adipogenesis. ROS not only damage cells but stimulate cell signaling trough oxidative protein modifications. ROS dependent signal induction may affect adipogenesis. S-glutathionylation, one of the oxidative protein modifications on cysteine by GSH adducts, can change protein function directly. S-glutathionylation is a reversible modification and eliminated by glutaredoxin(Glrx). However, little is known about S-glutathionylation in adipogenesis and obesity. In this study we examined roles of S-glutathionylation in adipogenesis and obesity by deletion of Glrx in 3T3L1 cells and mice. Hypothesis: S-glutathionylation affects adipogenesis and obesity. Methods: Body weight and fat mass of Glrx KO mice and littermate control fed with normal chow was analyzed.
