ThecDNAofhumanDNApolymerase 6wascloned. ThecDNAhadalength of3.5kbandencoded aprotein of1107aminoacid residues with acalculated molecular massof124kDa.Northern blot analysis showedthat thecDNAhybridized toamRNAof3.4kb.Monoclonal andpolyclonal antibodies totheC-terminal 20residues specifically immunoblotted thehumanpol6catalytic polypeptide. A multiple sequencealignment was constructed. Thisshowedthathumanpol6isclosely related toyeastpol6 andtheherpesvirusDNA polymerases. Thelevels ofpol6messagewerefound tobeinduced concomitantly withDNApol6activity and DNA synthesis inserumrestimulated proliferating IMR90cultured cells. Thehumanpol6genewas localized tochromosome 19bySouthern blotting of EcoRI digested DNAfromapanel ofrodenVhuman cell hybrids.
