Effects of SSBH on Expression of TNF-α, MMP-1 and MMP-3 in collagen-induced Arthritis, a Preparation from an anti-arthritic Chinese Herbal Formula

[Objective] To discuss the effects of San Shui Bai Hu decoction (SSBH) on the expression of TNF-α, MMP-1 and MMP-3 in collagen-induced arthritis (CIA). [Method] 33 CIA rats were randomly processed in 3 groups. SSBH, leflunomide and sodium chloride were intragastrically administered respectively. After 30 days' treatment, the rats were sacrificed. In situ hybridization was performed on joint tissue sections to observe the expression of MMP-1 and MMP-3. Enzyme linked immunosorbent assay was performed to detect the serum amount of TNF-α. [Result] In SSBH group and LEF group, the serum amount of TNF-α was significantly lower compared with control (P＜0.05). In SSBH group, the mean stained area and the staining intensity for MMP-1 and MMP-3 were significantly weaker than control group (P＜0.05). [Conclusion] SSBH exerts an inhibiting effect on the expression of TNF-α, MMP-1 and MMP-3 in synovioblasts of CIA.
