Modulation of Aloe Polysaccharides on X-ray irradiated non-malignant cells in apoptosis and apoptotic regulating protein expression

AIM To study the modulation of Aloe Polysaccharides on the apoptosis and apoptotic regulating protein expression in X-ray irradiated non-malignant cells. METHODS The apoptosis was analyzed using flow cytometry. The level of apoptotic regulating protein expression was tested using Western blot. RESULTS A distinct DNA ladder happened in C Liver cells 72 h after irradiation. Pre-treatment of AP in the concentration of 25 or 50 mg·L -1 significantly decreased the DNA ladder. The apoptotic rate of C Liver in 6 h and 72 h after irradiation was decreased from 5.0%, 43.0% to 2.2% and 10.9%, respectively. Meanwhile, pre-treatment of AP down-regulated the expression of p53, Bad and Bax protein caused by irradiation and evidently up-regulated the expression of Bcl-2 and Survivin protein. On the other hand, The apoptotic rate of 293 cells in 6h after irradiation was decreased from 8.6% to 4.0%. CONCLUSION The inhibiting effect of AP on the apoptosis of non-malignant cells was at least partly caused by the modulation of apoptotic regulating protein expression.
