The present invention relates to activator compound of Peptidylarginine deiminase in epidermis 1 and/or 3 and application thereof.Especially, the present invention relates to the activation of Peptidylarginine deiminase (PAD) 1 and/or 3 in the epidermis of at least one activating agent by being used alone or in combination in cosmetic composition and/or pharmaceutical composition (i.e. caffeine, theophylline acetic acid and/or theobromine).The invention further relates to purposes of the foregoing active agents in cosmetics and/or therapeutic agent, either individually or in combination, to improve the barrier function of epidermis, prevention and/or the treatment symptom related to dry skin, or improve and/or promote the moisture-keeping function of cuticula.Finally, the present invention relates to the activity of PAD1 and/or PAD3 in increase epidermis.
