人参皂甙-Rh2对Ara-c诱导HL60细胞增殖抑制和凋亡作用的影响

Objective To investigate the effect of GS-Rh2 on anti-leukemia action of Ara-c. Methods Human myeloid leukemia cell (HL60) was cultured in virto and divided into three groups, Ara-c group was treated with Ara-c at the flnal concentration of 5.0~40.0 μg/ml, GS-Rh2 group was treated with GS-Rh2 at the final concentration of 4.0~32.0 μg/ml, the combination group was treated with GS-Rh2 and Ara-c of above concentration. 48 h later, the inhibition ratio (IR) of the three groups were evaluated by MTT assay respectively, the half maximal inhibitory concentration (IC50), combined index (CI), efficiency multiples and the ratio of combined effect to expectation combined effect (q) were calculated. Morphology of apoptosis were studied by inverted microscope and staining of Right-Giemsa. Results IC in Ara-c group and GS-Rh2 group were 32.0, 13.0 μg/L; IC50 of Ara-c in combination group was 14.5 μg/L, which was significantly lower than that in Ara-c group (P<0.001). CI in combination group was 0.76 and the efficiency multiples was 2.21, the value of q was 1.42, 1.39, 1.36, 1.21 at different Ara-c concentration of 5.0, 10.0, 20.0, 40.0 μg/ml respectively; Morphology of apoptosis cell was more obvious than the other two groups. Conclusions GS-Rh2 can enhance the efficacy of Ara-c by increasing the sensitivity of HL6O to Ara-c, reducing the dosage and side effects of Are-c. These results provide a new thought for clinical treatment of leukemia.
