Protein production procedures using antisenescence compounds

2008
A method of producing a TNFR fusion protein in cell culture comprising the steps of: culturing mammalian cells that contain a gene encoding the TNFR fusion protein, expressing said gene under cell culture conditions, in a cell culture medium that it comprises an antisenescence compound, in which the antisenescence compound is selected from the group consisting of incarnosine, acetylcarnosine, homocarnosine, anserine and beta alanine and combinations thereof and in which the antisenescence compound is present in the cell culture medium at a concentration of approximately half 5 mM and approximately 100 mM; and keep the culture in sufficient conditions and for a time to allow protein expression; in that the cell culture shows an improved cell culture characteristic that differs from a corresponding cell culture characteristic that would be observed under otherwise identical conditions in an otherwise identical medium lacking the antisenescence compound; in which the improved culture characteristic is selected from the group consisting of: increased titer, increased specific cell productivity, accumulation of aggregates of reduced high molecular weight, accumulation of reduced acid species and combinations thereof.
