Exercise-Induced Circulating Profiles and Ex-Vivo Leukocyte mRNA Expression of Inflammatory Cytokines/Chemokines in Healthy Children and Children with Type 1 Diabetes

1x10 6 fusion cells on days 3, 7, and 10 post tumor implantation. Tumor volume (a 2 x b, a = longest diameter, b = shortest diameter, in mm 3 ) and mouse survival were then recorded. Results: DN TGF-β R expression was detected in 4T1 but not DCs transfected with adeno-DN-TGF-β R. DN-TGF-β R was able to bind to TGF-b and inhibited its activity by lung mink cell inhibition assay. TGF-β is known to induce regulatory T cells (Treg), thus we further showed that DC/4T1+Adv-DN-TGF-β R fusion cells induced less CD4+CD25+ Foxp3+ Tregs than DC/4T1+Adv-lacZ fusion cells. In vivo, DC/4T1+Adv-DN-TGF-β R fusion vaccine showed enhanced protective and therapeutic efficacy compared to the efficacy of DC/4T1+Adv-lacZ fusion vaccine (P < 0.05). Conclusions: TGF-β blockade resulted in lower Tregs induction by DC vaccine and enhances antitumor immunity. Expression of DN-TGF-β R by tumor fusion partner is an effective strategy to enhance DC/tumor - based antitumor vaccines.
