Protective effects of TRPV1 in hepatocytes apoptosis treated with high fat

OBJECTIVE To investigate the alteration of transient receptor potential vanilloid 1( TRPV1) in hepatocytes treated with high fat and its role in this process. METHODS The hepatic cells( BRL- 3A) were divided into three groups: control group,250μmol·L-1triglyceride group and 500 μmol·L-1triglyceride group. Then,capsaicin and capsazepin were administrated into hepatocytes for further intervention. The expressions of TRPV1 and hepatocytes apoptosis were measured by Q- PCR and flow cytometer respectively. RESULTS The apoptotic ratio of hepatocytes was concentration- dependent to high fat and the TRPV1 mRNA expression was significantly increased at 500 μmol·L-1triglyceride. Moreover,capsaicin obviously reduced the apoptosis of hepatocytes induced by high fat via increasing TRPV1 expression,which was reversed by the application of capsazepin. CONCLUSION High fat can activate TRPV1 pathway in hepatocytes,and TRPV1 enforcement is able to alleviate the hepatocytes apoptosis.
