Insan lenfosit hücre kültüründe melatonin ve b-karotenin mitomisin C ile indüklenmiş kardeş kromatid deşişimi sıklışı üzerine in vitro etkileri

Saglikli 11 bireyin periferik kan lenfosit kulturlerinden Fluorescence Plus Giemsa (FPG) teknigi modifiye edilerek kardes kromatid degisimi degerleri tespit edilmistir. Melatoninin ve I²-karotenin (beta-karoten) tek baslarina eklendikleri kulturlerle ve madde icermeyen kontrolle karsilastirildiginda benzer KKD degerlerine sahip olduklari, melatoninin ve –karotenin birlikte kulturlere eklenmeleri sonucu, madde icermeyen kontrol ve tek baslarina kullanildiklari kulturlerin degerleri ile karsilastirildiginda KKD degerlerinde farklilik olusturmadiklari gozlenmistir. MMC, beklenen KKD deserini arttirma etkisini gostermistir. Melatonin ve I²-karoten tek baslarina ve birlikte kullanildiklari durumda MMC.nin KKD.ni indukleyen etkisini azaltmistir. MMC.nin tek basina kullanimindaki sonucla karsilastirildiginda melatonin, I²-karoten ve MMC.nin birlikte kullanildigi kulturde KKD orani anlamli duzeyde azalmistir. MMC.nin KKD olusumunu indukleyen etkisi karsisinda I²-karoten, KKD degerlerini melatonine kiyasla daha fazla azaltmis ve melatonin ile karsilastirildiginda daha guclu antimutajenik etki gosterdigi saptanmistir. Anahtar Sozcukler: kardes kromatid degisimi, melatonin, beta-karoten, mitomisin C, genotoksik etki Abstract The in vitro effect of melatonin and -carotene on sister chromatid exchange frequency in mitomycin C-induced human blood lymphocytes The aim of this study was to investigate the in vitro effects of melatonin, I²-carotene (known as antioxidants) and Mitomycin C (MMC) (known as a genotoxic agent) separately and together on sister chromatid exchance (SCE) in human blood lymphocyte cultures. A total of 11 healthy human donors have been included in this research and their peripheral blood culture cells were processed by modified fluorescence plus Giemsamethod to investigate sister chromatid Exchange frequency. The results indicate that melatonin and I²-carotene (beta-carotene) alone and compared to untreated controls have similar incidance of SCE. Melatonin and beta-carotene together in the cell culture did not exhibit significant change in the frequency of SCE compared to the cultures they were alone and untreated controls. The lymphocytes, which were treated with mitomycin C (MMC), showed the expected increased incidence of SCE. Melatonin and I’-carotene used together and alone decreased the MMC induced frequency of SCE. Melatonin, I²-carotene and MMC together have significant decrease of SCE frequency when compared to MMC alone. We observed that, beta-carotene decreases the frequency of SCE more than melatonin. So, we concluded that beta-carotene showed stronger antimutagenic effect than melatonin according to the frequency of sister chromatid exchange.
