The invention relates to a tetraploid chemical induction method applicable to short-necked clams. The method comprises the main steps that after short-necked parent clams are subjected to sufficient maturity promoting and spawning induction, the short-necked parent clams discharge gametes; non-polluted high-maturity sperm eggs are selected, and fertilized eggs are obtained through artificial insemination; cytochalasin CB is added for treatment 13 min after insemination, the treatment concentration is 0.5 mg/L, release of a first polarbody is restrained, and the eggs are washed 19 min after insemination to remove CB; the fertilized eggs are placed under appropriate conditions for development, and tetraploid short-necked clam embryos are obtained; seawater adopted in the treatment process and the development process is sand filter sea water, the salinity is 24-26, and the temperature is 24.8-25 DEG C. The development synchronism of the fertilized eggs is effectively improved, the tetraploid induction rate is remarkably improved, and the inductivity detected by the flow cytometry is stabilized to be 50% or above.
