Application of the Virtual Cancer Patient Engine (VCPE) for improving oncological treatment desig

2004
692 Background Physicians are limited in their capacity to individually patient-tailor the safest and most efficient treatments in terms of drug and schedule choices, partly due to individual response variability. We have developed a computerized mathematical tool (VCPE), which was successful in retrieving In Vivo experiments. In the present work we retrospectively validated the tool and studied its ability to suggest improved Doxorubicin treatments. Methods VCPE, a combined platform of computerized mathematical models, simulates short/long-term cellular dynamics of specific patients undergoing drug therapy. It integrates patient hematopoiesis and tumor progression, from avascular growth through angiogenesis and metastasis, with a pharmacokinetics model accounting for drug Absorption, Diffusion, Metabolism and Excretion, and a pharmacodynamics model quantifying drug effects on several target tissues. Results VCPE retrospectively retrieved results of chemotherapy and biologic drug treatments: Tumor Growth-...
