Absrraet-2S-1-Cyano-2-hYd~oxy-3.4-epithiobutanes (erythro and thl'l!ol isolated from th~ seed .of Crambe abyssinica. were fed to groups of six weanling rats at levels of O. 75. 150 or 300 ppm In the ~Iet for 9{) days. The higher dose groups showed poor growth and incr~ seru~ levels o~ alkaline phosphatase. total bilirubin. gJutamic-oxaloacetic transaminase. gJutamlc-pyru~lc t~sammase and ornithine carbamyl transferase. The severity and occurrence ~f renal and hepatl~ lesIons ~ere dose: dependent Renal alterations consisted .of hYl'l7rtro!'hy of proxImal t~buJar epIthelial cells WIth .p~oml nent karyomegaly. Hepatic lesions consIsted pnIl?anly o.f mega.locytosls of the hepatocyt~ and bile duct hyperplasia with disruption of the normal hepatic architecture. Half of the rats In the hlgh.dose group had karyomegaly of pancreatic acinar cells.
