Test of Dendritic Cells Loaded with Apoptosis of Brain Glioma Cells Induced by Heat-shock and Activated T cells In Vitro

Objective To prepare dendritic cell (DC) loaded with apoptosis of brain glioma cell induced by heat-shock and discuss its’ activity agaist tumor. Methods Human peripheral blood-derived immature DCs were induced. DCs were loaded with apoptosis of brain glioma cells induced by heat-shock. Its phenotype was detected by Fluorescence double labeling flow cytometry and the activity of cytotoxic T lymphocytes(CTLs) were detected by MTT colorimetric assay. Results DCs had fine activable effect on CTL. As far as the function of killing brain glioma is concerned, CTLs activated by DCs loaded with heat-shock brain glioma cell were significantly higher than DCs loaded with freeze-thaw-cracking brain glioma cells(P0.05), with a higher effector-target ratio resulting in higher killing. Conclusion DCs loaded with heat-shock glioma cells are safe and effective tumor vaccine in clinic application.
