The effect of Y-12, 141, levamisole (LMS) and D-penicillamine (D-Pc) on delayed type hypersensitivity (DTH) in mice was investigated. Four strains of mice were compared for susceptibility to sensitization with methylated human serum albumin used as the antigen and ranked ICR>BALB/c>C3H/He>C57BL/6. Y-12, 141 (100 mg/kg, p.o.), LMS (30 mg/kg, p.o.) and D-Pc (300 mg/kg, p.o.) showed a significant enhancement of the DTH in the naturally low responder mice (C57BL/6) to the antigen. These test agents also restored the artificially suppressed DTH in C3H/He mice treated with cyclophosphamide, hydrocortisone or carrageenan. In contrast, when naturally high responder mice (ICR and BALB/c) were used, D-Pc had tendency to suppress the DTH, and Y-12, 141 (100 mg/kg, p.o.) and LMS (10 mg/kg, p.o.) significantly inhibited it. Both agents, however, had no effect on the DTH in moderate responder mice (C3H/He) . These results suggest that Y-12, 141 showed an immunomodulating activity on cell-mediated immunity.
